Should I pick up gear with Mp5 and/or spirit for regen?
Pick up gear that as it if it's a general upgrade, but don't seek it actively.
And next, should I or shouldn't I gem for pure regen?
No, unless you really have a HUGE mana issue, otherwise, if you really feel like you need more regen, gem into SP+mp5/int/spirit, but try to avoid pure regen gems.
I also read that a large part from your regen should come from your intellect. Does this mean I need to socket for intellect a lot?
If mana is an issue, using SP+int in your yellow sockets is a possible solution.
And then about troughput, Haste, crit and spellpower are all good I read. But how do I know what I need more? And how do I decide whether I put intel, crit or Haste in yellow gem slots?
You need SP over anything else. You can use SP+crit or SP+haste in yellow sockets with a socket bonus worth activating, but in most cases, pure SP is the way to go for Disc.
Okay, long and confusing story. To summarize that:
- How do you get enough longevity? (Mp5 vs Intel)
Mostly from Intellect, mp5 and spirit don't hurt either, but shouldn't be seeked actively, gemmed or enchanted for.
- How should you fill up gem slots?
Runed Cardinal Rubies all the way.
- How should you select gear? (haste vs crit vs spirit vs Mp5)
SP>Crit%>Haste>Intellect>mp5=Spirit

Well...You do need one blue to keep the meta gem active! Mp5 and spirit are both fairly sub-par stats for disc, so go with whichever is cheapest tbh. ^^

If you have extra Mana @ the end of a raid boss, you have too much int. (so start grabbing SP> Crit>Haste.) If not, get more int.
I'd say if you have extra mana at the the end of raid boss with a less than average set of players. If you gem yourself solely for the elite players, you're going to be in big trouble if you ever run with a raid that is less accomplished.

Its very difficult to prove there's a difference.
Someone with a smaller effective mana pool will have a higher throughput, and thus need to heal less often, potentially increasing their effective mana pool through increased oo5sr time, and needing to cast fewer spells, thus draining their pool less.

Well, one should gem his gear according to the situation he most often finds himself in...
...and use enough appropriate gems to activate his meta, obviously.
