Thats precisely why it is a threat loss to try and incorperate DnD into your rotation, it kills the rotation.
Doing your rotation correctly, you can get a constant flow of strikes bringing in a constant threat boost, DS for DRes, HS when available.
Cast DnD, and you lose your rotation, and while DnD puts out more threat, it puts it out slightly more gradually than hitting HS after HS.
The set bonus makes DnD that much better an opener (ERW), and much better trash threat holder, but costs to many runes to try and incorperate it into a single boss fight.

i'd glyph into dnd just to make your Aoe threat that much better. then use a normal single target rotation when fighting a single boss. glyph of disease imo is a convience/aoe glyph. but if your aoe is gonna be covered with a better dnd then that just leaves you with convience.
blood has no issues with single target threat with or without set bonuses. so worrying about single target threat is a non-issue imo. but it's your char...i just think that glyphing dnd and wearing the 2pt10 would fix any aoe issues blood normally has.

The glyph on it's own covers pretty well for trash pulls in my experience; any further trouble managing packs usually comes down to retarded DPS.
I will also likely be dumping the DnD glyph once I get 2pcT10 and re-glyphing maybe RS, seeing as my other personal preference of using the disease glyph does weaken my single-target threat a small degree.
