I use Vuhdo with mouseovers. I've never been particularly convinced that the popular "clique" method has any advantages.
The only thing I do in particular is set the frames to react to left and right click with "target" and "menu" (respectively) since, by default, left-click will be a heal of some sort but presumably you'll want it to select the character like normal frames do.

I got mouseovers mixed with clicks.
Got ctrl+click for picking the target/menu.
I got most instants on clicks (and regrowth), nourish/healing touch/swiftmend on mouseover.

I'm personally a user of mouse-over macro's, grid, and clique. I only use clique to bind my middle mouse button to WG, and a side button for NS+HT. Otherwise my mouse-overs are bound to keys. For hot tracking I use GridIndicatorCornerPlus and GridStatusHots. Very clean small colored dots that you can set up to represent whatever you want, including progressively different colors to indicate how many stacks of LB are present, and color coded "timers" to show when hots are about to fall off.
Easily the hardest part of switching to different unitframes if you are using mouse-overs is having a very good way of tracking hots. Grid doesn't do this on its own without those extra addons, but it works beautifully after you set it up. I cant say that I've tried vuhdo, just figured I'd give my 2c of what works for me. Currently 2/7 hm Firelands.
